报告人简介:Jianbin Ruan博士,美国康涅狄格大学健康中心免疫学系助理教授。博士毕业于中国科学技术大学,曾在哈佛医学院及波士顿儿童医院从事博士后研究。主要研究天然免疫信号传导及炎症性细胞死亡的结构机制,重点关注gasdermin成孔与调控。
报告摘要:Innate immune cells detect infection and cellular damage and respond by initiatinginflammatory signaling pathways that can culminate in lytic cell death. A central unresolvedquestion has been how molecular sensing of danger is converted into physical disruption ofcellular membranes and release of inflammatory mediators. In this talk, I will present ourwork defining the structural and mechanistic basis of gasdermin-mediated pyroptosis, a keyinflammatory cell death pathway. Using biochemical reconstitution and cryo-electronmicroscopy, we determined high-resolution structures of multiple gasdermin pores, revealinghow these proteins assemble into large B-barrel complexes that perforate membranes. I willalso discuss how functional diversity within the gasdermin family is controlled, focusing onisoform-specific regulation of gasdermin B. Finally, I will highlight how bacterial pathogenssubvert this pathway, using Shigella as an example of how this host cell death machinery istargeted to evade immune defense. Together, these studies establish a structural frameworklinking innate immune sensing to execution and provide insight into how inflammatory celldeath is regulated in health and disease.
