报告摘要:
Biocatalytic transformations are increasingly important for green synthesis, but their integration with photocatalysis is limited by enzyme degradation from reactive oxygen species (ROS). We developed core–shell polymer micelles and vesicles with embedded organic AIE luminophores that act as visible-light photocatalysts[1]. These colloids efficiently regenerate NAD+ from NADH while protecting enzymes from ROS, enabling stable, recyclable photobiocatalysis under aerobic conditions with potential for industrial application.
Beyond synthesis, NADH/NAD+ cofactors are central to cellular metabolism, and their disruption can impair cancer cell viability. Remarkably, our AIE polymer colloids catalyze NADH/NAD+ interconversion even under oxygen-deficient conditions, bypassing common electron acceptors. In vitro and in vivo studies show that under hypoxia, these colloids induce severe NAD+/NADH imbalance, suppressing oxidative phosphorylation and glycolysis, and triggering energy crisis in 4T1 cancer cells and tumors[2]. These findings highlight their promise for hypoxia-targeted cancer phototherapy.
