新加坡南洋理工大学王东安博士:Living Hyaline Cartilage Graft (LhCG) for Cartilage Repair and Anti-Arthritis Drug Evaluation

In this study, we have developed a continuous methodology to directly set up a scaffold-free macro-scaled three-dimensional (3D)living hyaline cartilage graft (LhCG) with the aid of a biomaterial-based interim scaffolding system. The practical performance of LhCG is evaluated in the knees of minipig model with full-thickness chondral defects for 6 months. Hyaline cartilage based neo-tissue fulfills the desired in situ reconstruction. LhCG is also employed as an engineered biomimetic/pathological tissue platform for anti-arthritis drug evaluation in vitro. Osteoarthritis(OA) and rheumatoid arthritis (RA) disease modelsare created with LhCGby replicating the inflammatory environment of an OA/RA joint via co-culturing LhCG with lipopolysaccharide (LPS)-activated macrophages as well as inflammatory synovial fibroblasts,after which the accuracy of this model for in vitro drug-testing was validated using a popularly applied nonsteroidal anti-inflammatory drug (NSAID) – Celecoxib®. The results suggest that this new OA/RA model is able to adequately mimic the native OA/RA cartilage and is suitable to be used as an in vitro model for predicting native cartilage response to drug treatment.

王东安博士主要从事生物材料、组织工程与再生医学方面的科研工作。迄今,作为主要作者,王博士共发表研究论文及会议论文>100篇,包括 >70篇学术期刊论文,2篇(章)专著论文,>20篇(次)邀请报告,>40篇(次)会议报告与摘要,并拥有发明专利若干。其中学术期刊论文包括发表在《自然-材料学》,《生物技术趋势》,《高级药物传输综述》,《高级功能材料》以及《生物材料》等国际权威杂志的文章。它们已被累计引用>1100次(ISI);部分论文被《科学》,《自然-材料学》,《化工技术》(英国皇家化学会),《组织工程》,《材料化学杂志》等刊物社论报道并予以好评;另外部分论文则被《高级药物传输综述》,《药物研究》,《材料化学杂志》,及《生物技术进展》等杂志采用为封面文章发表。王博士于2010年发表在《欧洲药物科学杂志》的一篇论文,荣获欧洲药物科学联盟颁发的2011年度最佳论文奖。
王博士先后应邀担任《高级药物传输综述》(2010年第7-8期)及《药物研究》(2011年第6期)杂志的主题期卷客座编辑。此外,王博士还应邀担任40多种国际研究性杂志的审稿人,以及包括新加坡国家科研基金会(NRF),新加坡卫生部(MoH)和新加坡科技研究局(A*Star)等新加坡国家级科研基金的经费审稿人。
